Assessment of circulating extracellular vesicles from calorie‐restricted mice and humans in ischaemic injury models

Calorie restriction (CR) and fasting affect lifespan, disease susceptibility response to acute injury across multiple animal models, including ischaemic injuries such as myocardial infarction or kidney hypoxia. The cargo function of circulating extracellular vesicles (EV) respond changes in host physiology, exercise, injury, other interventions. Thus, we hypothesised that EVs induced following CR may reflect some the beneficial properties itself. In a pilot study, were isolated from mice 21 days 30 % CR, eight human donors after 72 h water-only fasting. EV size, concentration morphology profiled by NTA, western blot cryoEM, their was assessed using assays related diseases. We found post-fasting samples better protected cardiac cells hypoxia/reperfusion (H/R) compared pre-fasting EVs. However, there no difference when used treat H/R-injured epithelial cells. Post-fasting derived slowed rate fibroblast migration slightly reduced macrophage inflammatory gene expression Lastly, miRNA cargos pre- serum significant small number miRNAs. conclude appears influence function, with varied effects worthy further exploration.
